Isolation and purification of esterase from green peas

The isolation and purification of esterase from green peas involves extracting soluble proteins from homogenized green peas, precipitating esterase with ammonium sulfate, dialyzing to remove excess salts, and finally purifying through ion exchange chromatography to obtain a highly concentrated and pure esterase enzyme.

Goals and Objectives:
  • Understand the process of protein extraction and purification from plant sources.
  • Apply techniques such as precipitation with ammonium sulfate, dialysis, and ion exchange chromatography for enzyme purification.
  • Obtain a highly concentrated and pure esterase enzyme suitable for further study.
Expected Outcomes:
  • Successful extraction of soluble proteins from green peas.
  • Precipitation of esterase enzyme with high yield using ammonium sulfate.
  • Removal of excess salts and impurities through dialysis.
  • Purification of esterase enzyme to achieve high concentration and purity.
  • Characterization of purified esterase enzyme.
Special Feature:
  • Utilization of ion exchange chromatography for the purification of esterase enzyme ensures high specificity and efficiency in obtaining a pure enzyme preparation.
Requirements from Participants:
  • Understanding of basic principles of biochemistry and protein structure/function.
  • Adherence to safety protocols during experimental procedures.
  • Documentation of experimental procedures and results in accordance with guidelines.
